Polymorphisms of estrogen receptors, ERα and GPR30: association with breast cancer susceptibility and prognosis
نویسندگان
چکیده
The purpose of this study was to clarify the roles of polymorphisms from the classical nuclear estrogen receptor ESR1 and from the recently described estrogen receptor coupled to G proteins GPR30 [1], in breast cancer susceptibility and prognosis. Three single nucleotide polymorphisms (SNPs), rs2234693 and rs9340799 from ESR1 and rs3808350 from GPR30 were genotyped in 260 breast cancer patients and 259 controls. SNPs were analyzed by PCR-RFLP and by real-time PCR with TaqMan probes. Genotypes were correlated with established breast cancer prognostic markers. For rs9340799, our results showed a significant association between A allele and breast cancer susceptibility, particularly for homozygous (OR-7.33, 95%CI, 4.3-12.6; p < 0.0005). The occurrence of polymorphisms rs2234693 and rs3808350 did not differ between breast cancer patients and controls. However, for rs2234693, CC genotype was significantly associated with higher (G2/G3) tumor grade (p < 0,05; OR-1.01, 95%CI, 1.01-4.98) and in postmenopausal women, the TT variant was associated with lower (G1) tumor grade (p = 0,02, OR-1.9, 95% CI, 1.09-3.45). No significant association was found with the presence of estrogen receptors or with HER2 overexpression in tumor samples. In conclusion, our work confirms the role of ESR1 polymorphisms in breast cancer: rs9340799 in breast cancer susceptibility and rs2234693 in breast cancer prognosis. For GPR30 SNP rs3808350, none association was found.

The Cell Surface Estrogen Receptor, G Protein- Coupled Receptor 30 (GPR30), is Markedly Down Regulated During Breast Tumorigenesis
BACKGROUND GPR30 is a cell surface estrogen receptor that has been shown to mediate a number of non-genomic rapid effects of estrogen and appear to balance the signaling of estrogen and growth factors. In addition, progestins appear to use GPR30 for their actions. Therefore, GPR30 could play a critical role in hormonal regulation of breast epithelial cell integrity. Deregulation of the events m...

Background: Breast cancer is the most common female malignancy world wide and in Iran as well and is the second cause of death due to malignancy after lung cancer. Varieties of factors such as estrogen and progesterone receptors, and axillary lymph node involvement may influence the prognosis and therapeutic approach. However, mutation in HER2 gene may also affect the prognosis. In this study...
متن کاملGPR30 regulates the EGFR-Akt cascade and predicts lower survival in patients with ovarian cancer
UNLABELLED OBJECTIVES G protein-coupled receptor 30 (GPR30) is a 7-transmembrane estrogen receptor that functions alongside traditional estrogen receptors to regulate the cellular responses to estrogen. Recent studies suggest that GPR30 expression is associated with a poor prognosis, and that this is due to the GPR30-mediated transactivation of the EGFR in breast cancer. However, the biologi...
متن کاملP-238: Lack of Association of Estrogen Receptor Polymorphisms with Male Infertility
Background: Estrogen is recognized as one of the significant regulator of spermatogenesis. Estrogens are synthesized in the male reproductive system (sertoli and leydig cells). Estrogen function is mediated by estrogen receptors (ER-α or ER-β). Some studies have suggested an association between single nucleotide polymorphisms (SNPs) rs2234693 (ESR1 pvuII C>T), rs1801132 (ESR1 325 C->G) of ERα g...
متن کاملAssociation of P53 (+16ins-Arg) Haplotype with the Increased Susceptibility to Breast Cancer in Iranian-Azeri Women
Background:Many case-control investigations have showed the correlation of TP53 gene polymorphisms with the risk of breast cancer. However, the findings are not consistent. It has been suggested that the investigation of P53 genotype combinations and haplotypes may be more helpful than the detection of single polymorphisms. In the present study, we investigate...
